I believe the game your referencing is a fan game. No Sonic game on the Genesis supports more than 2 players.

Do your options match your rom types? I.E split sets looking for split roms? Sounds like it is just not finding the files.

If they map to the cross hair, make sure they are going full up and full down. If they do then your only option is joy2keys, if it does not reset calibration.
