Have you enabled joystick control in the mame32 options?. When you open Mame32 go to the toolbar at the top and select 'options' then 'default game options' then 'controllers'. Tick the joystick tab. Should be ok after that.
However, there are a few controllers that have issues with mame.
If the above doesn't work then your best bet would be to download Joy2Key (http://www.electracode.com/4/joy2key/JoyToKey%20English%20Version.htm). What you do is use J2K to assign your controller button presses to keyboard keys. Then you assign those keyboard keys in mame and voila!.
Hope that helps.

With any advice about options in mame32 you can transfer it to commandline mame.
Look for the mame.ini in your mame folder and all the same options are in there. Simply use '1' to turn functions on and '0' to turn them off.
